However, once reconstituted, stability decreases significantly; reconstituted solution must be used within 2-4 weeks.

Choosing the Reconstitution Diluent

Two options: Sterile Sodium Chloride (0.9% injectable solution, also called normal saline) is standard and stable long-term. Bacteriostatic Water (sterile water with 0.9% benzyl alcohol preservative) has enhanced antimicrobial properties for multi-dose vials stored 2-4 weeks. Either is acceptable; sodium chloride is more common and slightly more stable.

Calculating Reconstitution Volume

Determine desired concentration. Standard: 1-2 mg/mL. If using 5 mg vial, add 5 mL diluent for 1 mg/mL or 2.5 mL for 2 mg/mL. Example: 10 mg vial = 10 mL diluent for 1 mg/mL. More concentrated solutions (2-3 mg/mL) require smaller injection volumes (easier for frequent use); less concentrated (0.5 mg/mL) are easier to measure precisely but require larger volumes.

Reconstitution Technique Step-by-Step

1) Prepare sterile workspace (wipe countertop with 70% alcohol). 2) Swab vial top with 70% alcohol prep, let dry 30 seconds. 3) Draw diluent into sterile syringe (amount calculated above). 4) Pierce vial top with needle, inject diluent slowly and steadily against vial wall (not directly into powder—minimizes foaming). 5) Remove syringe, gently swirl vial 30-60 seconds until powder fully dissolves (do NOT shake vigorously).

Signs of Proper Reconstitution

Solution should be clear, colorless or very slightly yellowish (not cloudy, not discolored). If cloudy or discolored, do not use (protein denaturation). Gentle swirling should dissolve powder within 1-2 minutes; if powder remains after 5 minutes, discard (failed reconstitution, possible contamination). Visual inspection is crucial.

Sterile Technique and Contamination Prevention

Use sterile technique throughout: sterile vial (sealed when received), sterile syringe/needle, sterile diluent, clean hands, swabbed surfaces. Never touch needle tip or vial opening. Use new sterile needle for each vial access. If reconstituting for multi-dose use, use sterile syringe cap between withdrawals. Storage of Reconstituted Orexin-A

Do NOT freeze (freezing denatures peptides, causes precipitation, and makes solution non-viable). Refrigerated, reconstituted Orexin-A is stable 2-4 weeks depending on diluent (bacteriostatic water extends to 3-4 weeks; normal saline ~2 weeks).

Multi-Dose Vial Management

If reconstituting for multiple doses (weekly/multi-week use), use bacteriostatic water and store refrigerated in darkness. Label vial with reconstitution date. Strict sterile technique on each withdrawal is critical: use new needle for each draw, swab septum with alcohol before each entry, don't leave septum exposed. Discard after 3-4 weeks even if solution remains.

Single-Dose Reconstitution

For immediate single-dose use, any diluent is acceptable. Prepare, inject immediately (or within hours), discard remainder. This minimizes contamination risk and eliminates storage concerns. Many users prefer single-dose preparation daily/weekly over multi-dose vials.

Troubleshooting Reconstitution Problems

Cloudy/precipitated solution: discard (denatured protein). Powder won't dissolve: verify diluent is sterile and at room temperature; if still won't dissolve, discard. Excessive foaming: inject more slowly or let foam settle 30 minutes before use. Discoloration: discard immediately (bacterial contamination or oxidation).

Shelf-Life Comparison: Powder vs. Reconstituted
